Description

Your impurity limit is not your pH limit.

Procaine hydrochloride, 20 mg/mL in water, has a pH of 5.83. Let 2 % of it hydrolyse and the pH is 4.66. The API barely changed. Its degradant, 4-aminobenzoic acid, took control of the pH from 11 µM, 0.0077 % of the API, far below any impurity limit.

The pH of a solution is not the pKa of the API. It is the one value at which every equilibrium in the flask agrees: the API, its counter-ion, every impurity and degradant, the buffer and water. pH Assist Pro solves all of them at once.


HOW IT WORKS

  1. Type the API. The structure, molecular weight and every published pKa are read live from PubChem, each with its source. ChEMBL predictions are labelled as predictions.
  2. See its degradation products. Curated pharmacopoeial routes for common APIs; for any other API, hydrolysis and oxidation products are predicted from the structure and identified in PubChem.
  3. Get the pH. Mass and charge balance, Davies activity coefficients, pKa and Kw corrected for temperature — the pH a calibrated glass electrode reads, with the state of every component.
  4. Find the impurities that matter. Each one is removed in turn: the pH shift it causes and the critical level at which it would move your pH by your threshold.
  5. Decide. Sensitivity and model uncertainty, acid or base needed for a target pH, the calibration buffers to use, and a measured-vs-predicted check.
  6. File it. An A4 report you can print or save as PDF.

VERIFIED

Checked against the NIST / IUPAC primary pH standards: seven of seven within ±0.02 pH.

The one exception is shown openly,alcium hydroxide, where ion pairing matters.
